Chuck is lost in a daydream at the Nerd Herd desk. Listening to an old mix tape, he thinks back wistfully to his old girlfriend Jill Roberts, the love of his freshman year at Stanford. But the memory becomes bittersweet as he recalls that she dumped him after he was wrongfully accused of cheating and expelled from the school. The pain returns as he remembers the aftermath and the day he went AWOL on his Buy More lunch break to try to win her back. He drove straight from Burbank to Stanford only to learn that she was now dating Bryce Larkin, the roommate who framed him in the cheating scandal. Still rhapsodizing about Jill, even though she broke his heart and slept with his ex-friend, Chuck is interrupted by his Assistant Manager Emmett Milbarge, who sends Chuck on an offsite support call.

Chuck's offsite service call is to set up a computer network at a biotech conference. As he's finishing the installation, he notices his old girlfriend Jill stroll into the lobby. Chuck tries to hide from her, but she spots him and they reconnect. She's a PhD now and is giving a lecture on infectious diseases at the conference. Chuck, embarrassed to be working at Buy More, tells Jill he has his own successful computer company. Jill introduces Chuck to her boss, biochemical researcher Guy LaFleur. Chuck flashes on Lafleur, witnessing a montage of biological weapons of mass destruction.

Back at their underground spy headquarters, Chuck, Casey, and Sarah learn from General Beckman that LaFleur is suspected of developing WMDs for sale to the highest bidder. Casey and Sarah's assignment is to surveil LaFleur while Chuck "reconnects" with Jill to determine the extent of her involvement.

At the Buy More, sycophant Emmett has brought Big Mike a box of donuts. Mike can't resist but quickly begins choking. Emmett performs the Heimlich maneuver, spewing the doughy blockage toward Lester. The deadly donut incident over, Emmett decides all Buy More employees will need to undergo emergency preparedness training. The CPR section will be taught by none other than Captain Awesome.

Chuck prepares for his big night with Jill. Dressed in a sharp suit and driving a rented Ferrari, Chuck picks up Jill and takes her to a swanky restaurant (staffed by CIA agents). With difficulty, Chuck tries his best to forgive Jill for dumping him in favor of his best friend. She feels bad and wishes she could smooth things over. Afterward, as they wait for Chuck's car, Jeff and Lester pull up in a Nerd Herd car. They begin razzing Chuck, telling Jill that Chuck works the Nerd Herd just like them, making $12.50 an hour. Disgusted by Chuck's deception, Jill jumps in a taxi and disappears.

The next day, Chuck joins Casey and Sarah in a surveillance van as they listen in on Guy Lafleur and Sarah's conversations in their hotel. One of their listening devices fails and Chuck volunteers to go up to Jill's room to plant a new one. After coming clean that he's just a Nerd Herder at the Buy More - and that he's never really gotten over their break-up - Jill invites Chuck into her room. As he looks for a place to hide the bug, it becomes clear that Jill still has feelings for Chuck, too. They share a passionate kiss.

Chuck returns to the surveillance van and notices an assassin headed for Guy Lafleur's hotel room floor. Casey and Sarah bolt to intercept him, leaving Chuck alone in the van, where he listens in on Jill's phone call with a girlfriend - and inadvertently joins the conversation, freaking Jill out. Casey and Sarah race to head off the assassin, but they're too late. The assassin hurls Lafleur through his hotel window; his body lands with a thump on the roof of the van.

Casey blindfolds Jill and takes her to Castle (their spy headquarters beneath the Orange Orange) for questioning. She explains that Lafleur had discovered that their company was preparing a bio-weapon for sale. His presentation at the conference was to have exposed their research and prevented its sale. With Lafleur dead, Casey suggests that should give the presentation to smoke out the assassins who could ultimately lead them to the bioterrorist customers. Chuck protests that Jill shouldn't be used as bait; Sarah volunteers to go in disguise to give the presentation herself.

Back at Buy More, the day for the employees' CPR written test has come. Much to Emmett's delight, the timed test is clearly going to prove tough for Morgan, Lester, and Jeff. They squirm to sit near Chuck, who's knowledgeable in CPR.

As Sarah begins her speech, a mysterious leaves a metal canister under his chair and runs from the room. Sarah gives chase, leaving Casey inside. He discovers that the canister is a time-release weapon, emitting a deadly biological agent into the air - and spelling certain death for everyone inside. In desperation, Casey calls Chuck and tells him to find Jill, apologize (again) and see if she has an antidote.

Chuck rushes from the Buy More to Jill's hotel room and confesses once again, this time explaining that he's a CIA agent. Noting that he's accompanied by a posse of rescue personnel, Jill finally believes Chuck and they all rush to the hotel to rescue Casey and the room full of scientists. Jill injects Chuck with the virus to create an antiviral serum he can use to make an antidote for the others. He bravely enters the sealed-off ballroom, carrying a centrifuge kit which he'll use to save the others.

Sarah chases the assassin who poisoned the ballroom down through the bowels of the hotel. In a shootout, she kills him - only to discover that he's apparently a FULCRUM agent posing as CIA.

Inside the contaminated ballroom, Chuck receives instructions from Jill on how to create the antiviral serum using the centrifuge. But as he and Casey try to draw blood into a glass vial, it falls to the ground and breaks. They call to Jill, who will try to find another; with just ten minutes left before the poison kills everyone, the situation grows desperate. Chuck reasons that the antivirus might be able to be transmitted through saliva and painfully kisses Casey in a bid to save his life.

Back at Buy More, time is running out for the test takers. Morgan, Jeff, and Lester have no idea of how to complete the Heimlich maneuver. In a moment of inspiration, Morgan and Lester hatch a plan to have Jeff swallow a ball point pen cap and begin choking. Then they'll watch how Captain Awesome performs the maneuver. Jeff dutifully swallows the cap. As Awesome demonstrates how to perform the life-saving move, Morgan and Lester take notes. They successfully complete the exam just as time runs out.

Jill arrives with a replacement vial and syringe and immediately creates an antiviral serum from Chuck's blood. They inject all the victims and emerge to a hero's welcome. Completely impressed by this new side of Chuck, Jill grabs him and they share a lengthy kiss.

Back at Castle, a recovered Casey asks General Beckman how they should handle Jill, now that she knows Chuck's a spy. Beckman explains that they'll use her as bait to find the remaining FULCRUM agents - and that they're not to explain this to Chuck, for fear that he'll tip her off and she'll flee. Sarah disapproves but goes along with the plan.

Blissfully ignorant of the plan and back in love with the girl he lost years before, Chuck calls Jill and sets up a real date. Apparently you can go back.
